The transcription factor cKrox was originally identified as a protein that bound to a negative transcription regulatory element in the murine alpha1(I) collagen promoter. We recently reported the cloning and characterization of human cKrox (hcKrox). Overexpression of hcKrox in NIH3T3 fibroblasts efficiently repressed the promoters of the fibronectin and alpha1(I) collagen genes (70-90%) in transient transfection assays and suppressed the endogenous genes in hcKrox expressing permanent cell lines. We have now isolated genomic clones and cDNAs encoding two novel transcription factors related to hcKrox termed hcKrox-beta and hcKrox-gamma (the original clone is now referred to as hcKrox-alpha). Both contain three kruppel-like zinc-finger DNA binding motifs that are 71-78% identical to those of hcKrox-alpha. The NH(2)-terminus of all three proteins contains a POZ domain, a conserved 120 amino acid motif involved in transcriptional repression and protein dimerization. RT-PCR experiments demonstrate that all three hcKrox family members are expressed in foreskin and dermal fibroblasts. Transient transfection studies in NIH3T3 fibroblasts demonstrate that hcKrox-alpha -beta and -gamma, as well as the murine cKrox-beta homologue, LRF, suppress transcription driven by promoters for the alpha1(I) and alpha2(I) collagen, fibronectin and elastin genes. Electrophoretic mobility shift assays and coimmunoprecipitation studies suggest that homo- and heterodimerization occurs between cKrox family members. Dimer formation is influenced by amino acids in the NH(2)-terminal POZ domain and the Zn(+2)-finger region. Immunoprecipitation studies indicate that cKrox can form heterodimers in solution in the absence of DNA. Thus, a multi-gene family exists that can coordinately regulate several extracellular matrix genes and has the potential to form many heterodimeric transcription factors.

Honestly, The stats on the last podcast we're really discouraging because there we 20 or so people streaming the podcast! We always get criticised for making the podcast about us! When we go out of our way to get the community involved hardly anyone gets involved.
I'm not complaining just stating my own frustration. Episode 32 was ONLY for the community and only one person wanted to take part!
As for getting the UK involved our buddy XPETRI from the UK is in the chat room every time we go live. But i realize our Bread and Butter is the USA and our average audience is working between 2-4pm est on a weekday! Our West Coast friends would be eating lunch!

David - now I feel bad that I didn't tune in for the live podcast! I have been so zonked from work (and the nearly 100 degree temperatures), and I didn't think anyone wanted to hear me snoring on my cell phone. Don't be discouraged!
On a lighter note - I thought the exact same thing about A Wrinkle in Time the first time I read it. I really need to unearth my copy and start reading. Since this book is shorter, I'm planning to start closer to the discussion date, so I don't forget what I want to talk about.
Speaking of books, here's a question for you all (everybody) to ponder: We seem to have run out of Watership talk relatively quickly, and we also seem to have some crazy fast readers on here. We have a few "extra credit" books in the line-up. Is anyone interested in me assigning one extra book to each month? That way, we'd have an official book for everyone, and an extra book for those who want to talk more, read superfast, have no lives (ha!). Just kidding. :)

P.S. I know Catch-22 is already on the extra credit list, but maybe we should make it required for the month that The Third Policeman had (if we combine it with the Alice books).

I would suggest you grab a copy of the annotated Alice. You will never look at that story as a breazy read again. I've read the ALice books a number of times over the years!! I love that book, but it's like a fine wine, it needs to be savored.

As for extra credit books, I just finished The Time Traveler's Wife and I know I'm probably the last person to have read it, but my LOST QOTD has to do with that book and Island pregnancies. There may be an explanation for what happens to the Island moms in TTTW.
Good morning! So much is going on it seems! I read this and it makes my mind race... Latest Harry Potter movie and then next weekend is the release of the last book! 10 years of Harry coming to an end - I can hardly believe it! Then comicon is the following week (no, I'm not going, but I'm sure there will be updates online)... in the meantime, the new JJ project which totally surprised me. I went to see Transformers on Friday and the Bad Robot stole the show! I can't keep up with this ARG, but I'm still glad it is happening. ;)
4400! I've been a fan since Season 2 (much like Lost) I watched a marathon of the first season and was hooked to start in on the 2nd and so on. I love these characters and I'm so glad the show keeps coming back!

I know what you mean. I just got back from getting my tickets for the Harry Potter movie tonight. They've already pre sold over a 100 tickets and they've only been open for a half hour. And I've had the last book pre-ordered for months it comes out on my birthday.

Did you know that they are re broadcasting Jercho? They showed the first epsode last Friday and tomorrow at 8:00 they're showing the recap show they showed after it's hiatis. Then at 9:00 they're showing the next in line and following through to the end of the season.

CBS claims that if they don't get enough "TV" viewers that they won't renew it. I say they need to catch up with the times. I hope they're not stupid enough to cancel it because of the internet viewers.

For anyone considering it, Harry Potter and the Order of the Phoenix was really good. I highly recommend it.
As far as the book club goes, we do actually have two books a month in August and September. We could discuss one for the first two weeks, and then the second the last two weeks of each month. I hate to change things for July since we're already so far in. Let me know what you guys think.

Also, I meant to be more involved with the Book Club, but I've been so busy working and rereading the Harry Potter books in anticipation of the new book, that I haven't had time. OT: Sayidsgirl, did you and your daughter like Surf's Up? I found myself throughout the movie realizing that I had a constant grin on my face! I thought that it was a lot of fun and very positive, although I really like surfer movies of any kind anyway. But anything that can make this crabby old lady smile for 2 hours has got to be good. BTW, the movie's website has a lot of great things for kids to do on it too.

Finished Jericho! Cool show. I can't believe they were going to end it like they did. Anybody else pick up on this show this summer?
Twinkle: I watched the show when it originally aired. I know, the ending was such a cliffhanger. Hopefully, enough viewers will watch this summer and they'll pick it up.
Hubby and I plan to watch Jericho when they air on Fridays via DVR. We watched the first one and taped last night. I hope they won't stop airing them like NBC did Friday Night Lights and like ABC did LOST last summer. I'd prefer to watch them on TV than on my computer.

I just realized, did you say you're only reading only three books on vacay Memphish? You're slipping. :o) BTW, I think there is a version out now of A Brief History, with tons of graphics to aid in comprehension for non-physicists like us. And there is also another new good book by him with profuse illustrations as well. But Brief History is not so bad. I listened to the audio version in my car and did OK.
Agreed, it was a very stupid way to end Jericho like that originally. I'm glad that it will be back for some more info eventually. This inability of TV programmers to commit to finish of a serial show is pretty strange. It's almost as if the viewers can handle them better than the programmers/execs can.

Finally saw Harry Potter 5 last night, and all I can say is wow. It captured the essence of the book, unfortunately they left lots of book details out but that's the way it goes when you do a screenplay. The book has so much more in it, always, yet this movie did the story justice, if not the book.
There's my review, imho. I can't wait for the dvd. Probly Christmas, eh?
Finished reading "A Wrinkle in Time". This one will be harder to relate to Lost, I think. At least not as obvious as "Watership Down".

Saw HP in Imax 3D on Friday - and what a blast! I'm most likely going to see it on a plain old big screen this weekend. I agree with amused that the movie does stand well on its own... now I'm just holding my breath waiting for the last book. I'm so afraid of being spoiled that I won't even browse online in fear I'll come across something. ;)
amused: I saw HP this weekend, and I completely agree with you. Although it was definitely not a stand alone, in that the friend I saw it with was totally lost as she had not read the book. And I was a little disgruntled that one of my favorites from the book was but a blip in the movie.
As for Wrinkle...I have loads of notes to compare it to Lost. However the comparisons are certainly a different sort than Watership's were.

It's some great pictures and a review of the new Lost figures. I have them pre-ordered, so I'll post my own photos on my blog when I get them. They look amazing!
I also have to agree with everyone who has seen Harry Potter. I thought it was a great movie, and they did a really good job adapting the book. Book 5 is SO packed with stuff, it must have been quite a task to whittle it down enough. There were a few scenes that I wish had been in there, just because I love them so much, but I don't think their exclusion took away from the movie at all.
I can't wait for the last book! I'm also trying to avoid all spoilers - but it's hard not to read everything Harry Potter related online!
Harry Potter fans - mugglenet.com is a great SPOILER-FREE site if you want whet your appetite for the new book. They have a section on Book 7, where they have debunked rumors, things that are known for sure (I guess these could be considered spoilers, although they're pretty mild) and my favorite - loose ends. You're right Mugglenet is a great Harry Potter site and they strive not to have spoilers. I've been going to that site for years along with The Leaky Cauldron www.the-leaky-cauldron.org is a spoiler free site also and sometimes colaborate with Mugglenet.
The Order of the Phoenix movie just shows you what a good director can do. I'm glad he's doing the next movie too. I don't know about you guys but I hated The Prisoner of Azkaban movie that crazy director tried to turn it into "art". Just my opinion. Can't wait for Saturday for the last book it's going to be exciting and sad.Thanks for the links to the new figurines.

Sayid's girl - I agree about the Harry Potter movies. I like Prisoner of Azkaban okay, but to me, it feels like a Cliffs Notes version of the book, more than any of the others. I understand that they can't fit everything from a book into a movie, but that one feels very choppy to me.
Maven - how do you like 24? Which season are you on? Personally, I like seasons 1, 3 and 5 the best - 5 is an absolute masterpiece. Season 6 was pretty lame, and I'm hoping that things pick up next year! I got my husband hooked with the season 1 DVDs, and we've been watching ever since. :)

Melissa_lossa: We're almost through with season 1 of 24. We did watch the season that just ended and heard all the complaints about it being the lamest (too much office romance). We're glad we decided to watch it from the beginning...a lot of discs to go through!

maven - I think all of the season's are good, with the exception of the newest one. Mostly, they just get better as they go on.

On Harry Potter: Melissa, I agree with you. Although I think "Prisoner of Azkaban" was a bare bones version of the book, I didn't find it choppy. I was ok with most of the subplots that got left out of the movie, especially the house elf liberation stuff.
"Order of the Phoenix", however, was very choppy, very Cliffs Notes. There was so much more that not only could have been done but (I think) should have been done. After all, if they can take 3 hours each to tell the Lord of the Rings trilogy, they can take 3 hours to tell a Harry Potter book. So put in a 5 or 10minute intermission, but tell the story!

yeah, but Lord of the Rings was 3 films. Not 5. And they were all filmed at one time.
Sad thing with Potter, is they do one at a time and we basically are guaranteed 7, total.
Basically, my point is this. They probably figure that since they've now made 5 films, the last thing they want to do is make them 3 hours long. Some people who aren't fans of the books might be getting tired of them by now. Hahahaha. I dunno. Just playing devil's advocate.

also, keep in mind, the lord of the rings was probably more anticipated than any of the harry potter movies. I'd go to see the LOTR trilogy AGAIN at the theater. However, I doubt I could do the same for Harry Potter.

They had the Emmy nominations on the Today show this morning and according to the Emmys website the only nominations for the actors were Terry O'Quinn and Michael Emmerson for supporting actor. Of course the show itself was not nominated for a drama series.

I just woke up to the Emmy news. Oh, well...I'm thankful Terry and Michael got the nominations, at least! It's just so frustrating that TPTB for the Emmys just don't get the greatness and depth of LOST and nominate pure entertaining "fluff" like Gray's Anatomy (I watch that show, but it's so superficial and has no mythology and nuances like Lost). We did get some technical nominations...Jack Bender for directing, Single-camera editing -- both for TTLG, Sound editing and Writing for a Series.
it would seem i was right about richard maybe not being back on lost, the cbs exec behind nester's new show cane said they prolly wouldnt let him do guest spots on lost, but i imagine cane will get canceled before it becomes an issue, considering its on cbs and not a procedural crime drama or military crime drama ...:)
I don't get how you can be nominated for Best Writing and Best Directing, and not get nominated for Best Drama. I mean, come on!

Wow- the Emmy news is a bit of a shocker. I really thought that the last part of Season 3 was really good and was bound to get more Emmy nods- esp. in best drama. I also thought Elizabeth Mitchell (Juliet) would get a nomination. *rant*- I just don't get all the hype around Grey's Anatomy. I mean isn't it just a night time soap opera? The episodes I've watched are ok, but nothing new or original- I just don't see what everyone else sees in that show!- *end rant* It will be interesting to see how Heroes does next year in the nominations. See if they drop Heroes like a hot potato just like they did LOST.
Oh well- at least LOST has got one of the most dedicated fan bases out there. That has to count for something!
I love my kids- but am so glad school starts again in 3 weeks! woo hoo!

you know the theory behind cbs's high ratings and i heard it on here,but i forget who. Is the people that watch CBS are so old that they fall asleep during the first hour of primetime and just never change the channel. Probably a good theory
greys anatomy is so so predictable. I used to like it but it can't match nip/tuck or house at least for me since they all have the same shocking case/ intricate personal lives formula, interspersed with montages set to ultra hip estrogen charged alternative rock. Oh wait the estrogen charged montages are just grey's.
2costa: i'm with you there. grey's hasn't been good in a while. occasionally they'll have a good episode, but i gave up on the whole everybody's havin' sex with everyone else (within the 8 person group, that is).

Speaking of which, they are for sure making a new X-Files movie! It's written by Chris Carter and Frank Spotnitz and directed by Chris Carter. Hopefully, it won't suck like the first one.
I'm about halfway through Wrinkle in Time, but I'll have to take a break this weekend for Deathly Hallows. I'm also in the middle of a nasty battle for a raise at work. Everyone cross your fingers for me!

So vacation was very relaxing and I managed to read The Drawing of the Three (Dark Tower Pt. 2), Lost Horizon, and the first 6 chapters of A Brief History of Time and 2 non-LOST related books, though 1 of them gave me a blog post idea. More on those in days to come.

cattygirl - I finished Harry Potter Saturday night at about 4 a.m. Great stuff. I wished that the epilogue had touched on more characters, but other than that, I thought it was a perfect ending. How about you?
I was checking out the new posts over at Lost Ninja. They are only up to "Greatest Hits" so far, but he had an interesting theory on Charlie's swimming ability:
In Charlie's fourth Greatest Hits flashback, we find him as a boy being taught to swim by his father. His brother Liam is telling him not to jump, as his father will surely move. As Charlie steps nervously to the poolside, however, a voice can be heard in the background, yelling "Come on, let's go - Desmond!"
At the end of the episode, Desmond lies unconscious in the canoe with a list of the five key moments in Charlie's life resting in his pocket. Knowing the importance of Charlie being able to swim, is it possible that Desmond somehow travelled into Charlie's past and altered the event so that Charlie can swim in the present and be the saviour they all require?
Dennis, I remember hearing that theory about Desmond being at the pool when Greatest Hits aired here. I still can't figure out how TPTB are going to deal with Desmond "changing" the past. It seems to open up too many cans of worms.
Speaking of changing the past, anyone who has read Drawing of the 3, Roland changes the past of someone pretty significantly near the end of the book, does King manage to reconcile this at some point in the future? A simple "yes/no/yes, but it's not convincing" answer will suffice. And who else thinks that Charlie was based on Eddie Dean?

never was excited about the time travel idea but i DO like the idea of the flash forwards (in MY mind) being future shots of "cause and effect".
example: the first flash forward is a future that will be because of jack's ultimate decision-making on the island. However, Locke was told by "Walt" that he still has work to do. (i think that's the same "entity" that spoke to Eko and Ben, etc. Smokey, the island....whatever).
Anyway, Locke's work is to prevent certain futures and/or to CAUSE some. i think he saw that he couldn't stop Jack, so he's now off to alter the future another way.
So.......now, the next flash forward will be based on the next major decisions made. Etc. and so-on.
Now, THAT'S an idea I can wrap my mind around.

Maybe that related to the chip in claire, maybe it makes a person like a roving webcam? how are we supposed to think that it was radzinsky in the polar bear cave, even though it was a pearl dharma logo on the guy in the polar bear cave and radzinsky had a question mark for the pearl on his map. Kelvin had a swan logo...i liked the secret clue where they played the room 23 clip backwards and it said something like "Only fools are trapped by time and space" and the guy watching the door from its always sunny in phily was reading a breif history of time. I saw that back masked on you tube before but to have a lost site affirm what is being said is cool...
but another reason I'm not "all for it" is because I think Charlie is more of a hero if Desmond did NOT interfere.
Don't mind Desmon seeing the future so-to-speak and what-not. Just not a fan of the time travel.
However, I'm NOT 100% AGAINST it either. eh.
